Restriction Digestion Kit (teaching Kit)

• Single or Double digestion can be performed as two different types of Restriction enzymes are provided.
• Easy determination of Molecular weight of the digested DNA fragments with provided marker. Restriction endonucleases (restriction enzymes) also known as molecular scissors are essential tools in molecular biology because of their ability to recognize and cut specific DNA sequences. Restriction enzymes hydrolyses the backbone of DNA between deoxyribose and phosphate group. This leaves a phosphate group on the 5′ end and
  hydroxyl group on the 3′ end of both the strands.

Plasmid Purification Teaching Kit

The objective of this kit is to understand the basic procedures involved in the isolation process of plasmid DNA from bacteria etc.

This Kit is design for preparation of plasmid by Alkaline lysis Method. The plasmid DNA is preferentially purified from other cellular components such as genomic DNA and RNA. This kit is able to purify plasmids from Bacteria. The typical purification yield is depends upon the sample type and sample volume.

Background

Alkaline lysis is a method used in molecular biology , to isolate plasmid DNA or other cell components such as proteins by breaking the cells open. Bacteria containing the plasmid of interest is first grown, and then allowed to lyse with an alkaline lysis buffer consisting of a detergent sodium dodecyl sulfate (SDS) and a strong base sodium hydroxide. The detergent cleaves the phospholipid bilayer of membrane and the alkali denatures the proteins which are involved in maintaining the structure of the cell membrane. Through a series of steps involving agitation, precipitation, centrifugation, and the removal of supernatant, cellular debris is removed and the plasmid is isolated and purified.
